Natural Antioxidants: Therapeutic Advances in Inflammatory Disorders and Cancer

Oxidative stress, which disrupts the cell’s redox balance, is a key factor in the pathophysiology of many human diseases. Exposure of our body to various oxidants, pollutants, allergens, infectious agents, and xenobiotics results in oxidative stress. Increased oxidative stress generates excessive amounts of reactive oxygen and nitrogen species and lipid peroxidation-derived aldehydes, which not only damage DNA, proteins, and lipids but also amplify oxidative responses and inflammatory pathways. These pathways disturb redox homeostasis, metabolism, and immune balance, leading to the onset and progression of various inflammatory diseases such as cardiovascular diseases, asthma, chronic obstructive pulmonary disease, sepsis, arthritis, neurodegenerative disorders, metabolic syndrome, and multiple cancers. Current treatment approaches using steroidal anti-inflammatory drugs and chemotherapeutic drugs often show adverse side effects. Therefore, there is a need to develop safer and more effective strategies. In this context, natural antioxidants derived from plants, functional foods, and bioactive phytochemicals have emerged as promising candidates. These potential natural antioxidants can neutralize harmful free radicals, restore the redox and immune balance, and impact signaling pathways associated with inflammation and carcinogenesis. The accessibility, dietary benefits, and long-standing use of natural antioxidants in traditional medicine make them a valued resource.
This special issue provides a comprehensive platform for investigations into the latest developments in this dynamic field. We welcome contributions in the form of high-quality research articles, reviews, and perspectives that highlight the identification of novel antioxidants, mechanistic insights, preclinical studies, and translational research demonstrating how natural antioxidants regulate oxidative stress, inflammatory cascades, and tumorigenesis.
